This variant lies in the NF1 gene (transcript NM_001042492.3) at coding-DNA position 7847, where A is replaced by G; at the protein level this means replaces glutamine at residue 2616 with arginine — a missense variant. Submitter rationale: The p.Q2616R variant (also known as c.7847A>G), located in coding exon 53 of the NF1 gene, results from an A to G substitution at nucleotide position 7847. The glutamine at codon 2616 is replaced by arginine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. <span style="font-family:arial,sans-serif; font-size:9pt">To date, this alteration has been detected with an allele frequency of approximately 0.002% (greater than 55000 alleles tested) in our clinical cohort. Based on protein <span style="font-family:arial,sans-serif; font-size:9pt">sequence alignment, this amino acid position is highly conserved in available vertebrate species.
