NM_000051.4(ATM):c.4241C>G (p.Ser1414Cys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 4241, where C is replaced by G; at the protein level this means replaces serine at residue 1414 with cysteine — a missense variant. Submitter rationale: The p.S1414C variant (also known as c.4241C>G), located in coding exon 28 of the ATM gene, results from a C to G substitution at nucleotide position 4241. The serine at codon 1414 is replaced by cysteine, an amino acid with dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6487 samples (12974 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.002% (greater than 66000 alleles tested) in our clinical cohort. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of p.S1414C remains unclear.
