Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000059.4(BRCA2):c.2585A>G (p.Lys862Arg), citing LabCorp Variant Classification Summary - May 2015: Variant summary: BRCA2 c.2585A>G (p.Lys862Arg) results in a conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be tolerated. The variant was absent in 227224 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2585A>G has been observed in individual(s) affected with/suspected of Hereditary Breast And Ovarian Cancer Syndrome (e.g. Ha_2020, Kim_2020, Park_2021, Dorling_2021). The variant has also been reported in unaffected controls (e.g. Dorling_2021 and Momozawa_2018). These reports do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 33471991, 33078592, 31907386, 30287823, 34063308). ClinVar contains an entry for this variant (Variation ID: 231603). Based on the evidence outlined above, the variant was classified as uncertain significance.
