NM_004304.5(ALK):c.1981A>G (p.Asn661Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N661D variant (also known as c.1981A>G), located in coding exon 11 of the ALK gene, results from an A to G substitution at nucleotide position 1981. The asparagine at codon 661 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
