Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000251.1(MSH2):c.-263C>G, citing Ambry Variant Classification Scheme 2023: The c.-263C>G variant is located in the 5' untranslated region (5&rsquo; UTR) of the MSH2 gene. This variant results from a C to G substitution 263 bases upstream from the first translated codon. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project; however, this position was not covered in the ESP. To date, this alteration has been detected with an allele frequency of approximately 0.002% (greater than 65000 alleles tested) in our clinical cohort. This nucleotide position is not well conserved on limited sequence alignment. Since supporting evidence is limited at this time, the clinical significance of c.-263C>G remains unclear.
