NM_000051.4(ATM):c.719T>G (p.Leu240Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L240R variant (also known as c.719T>G), located in coding exon 6 of the ATM gene, results from a T to G substitution at nucleotide position 719. The leucine at codon 240 is replaced by arginine, an amino acid with dissimilar properties. This alteration has been detected in a cohort of 122 patients who underwent multi-gene panel testing for hereditary cancer after having previously tested negative for mutations in BRCA1 and BRCA2 (Yadav S et al. Fam Cancer, 2017 Jul;16:319-328). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
