Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.2395C>G (p.Gln799Glu), citing Ambry Variant Classification Scheme 2023: The p.Q799E variant (also known as c.2395C>G), located in coding exon 14 of the RAD50 gene, results from a C to G substitution at nucleotide position 2395. The glutamine at codon 799 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
