This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 3798, where C is replaced by A; at the protein level this means replaces serine at residue 1266 with arginine — a missense variant. Submitter rationale: This missense variant replaces serine with arginine at codon 1266 of the BRCA1 protein. Computational prediction is inconclusive for the variant impact on protein structure and function. To our knowledge, functional studies have not been reported for this variant. A multifactorial analysis has reported a likelihood ratio for pathogenicity based on health history of approximately 0.82 from the reported log(LR) of -0.085030213 (PMID: 31853058). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
