Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_001048174.2(MUTYH):c.920A>G (p.Asn307Ser), citing ACMG Guidelines, 2015: This missense variant replaces asparagine with serine at codon 335 of the MUTYH protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. In a large international case-control study, this variant was reported in 1/60466 breast cancer cases and 3/53461 unaffected controls (OR=0.295, 95%CI 0.031 to 2.833, p-value=0.347PMID: 33471991). This variant has been identified in 1/230582 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
