This variant lies in the RAD51D gene (transcript NM_002878.4) at coding-DNA position 164, where G is replaced by A; at the protein level this means replaces arginine at residue 55 with glutamine — a missense variant. Submitter rationale: The RAD51D c.164G>A (p.R55Q) variant has been reported in heterozygosity in at least one individual being evaluated for hereditary breast and ovarian cancer syndromes (PMID: 31159747). It was observed in 4/282618 chromosomes in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID 230266). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
