NM_000465.4(BARD1):c.221G>C (p.Cys74Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C74S variant (also known as c.221G>C), located in coding exon 3 of the BARD1 gene, results from a G to C substitution at nucleotide position 221. The cysteine at codon 74 is replaced by serine, an amino acid with dissimilar properties. This alteration was identified in an individual diagnosed with breast cancer (Weber-Lassalle N et al. Breast Cancer Res, 2019 Apr;21:55). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
