NM_024675.4(PALB2):c.2325A>C (p.Gln775His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q775H variant (also known as c.2325A>C), located in coding exon 5 of the PALB2 gene, results from an A to C substitution at nucleotide position 2325. The glutamine at codon 775 is replaced by histidine, an amino acid with highly similar properties. This alteration was detected in 1/5589 German BRCA1/2-negative probands with breast cancer (Hauke J et al. Cancer Med, 2018 04;7:1349-1358). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
