Submitter rationale: The CHEK2 c.132C>T (p.S44=) variant has been reported in at least two individuals with hereditary breast/ovarian cancer syndrome (PMID: 34299313). It was observed in 7/113516 chromosomes of the Non-Finnish European subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID 230091). In silico tools suggest that the variant may have an impact on splicing, though these predictions have not been confirmed by functional studies.
