Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000051.4(ATM):c.931A>G (p.Ile311Val), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 931, where A is replaced by G; at the protein level this means replaces isoleucine at residue 311 with valine — a missense variant. Submitter rationale: Variant summary: ATM c.931A>G (p.Ile311Val) results in a conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be tolerated. The variant allele was found at a frequency of 8e-06 in 250896 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.931A>G has been observed in an HBOC individual with limited information (lack of co-occurrence and cosegregation data) (Tavera-Tapia_2017). These report(s) do not provide unequivocal conclusions about association of the variant with Breast Cancer. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 27913932). ClinVar contains an entry for this variant (Variation ID: 229990). Based on the evidence outlined above, the variant was classified as uncertain significance.
