This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 931, where A is replaced by G; at the protein level this means replaces isoleucine at residue 311 with valine — a missense variant. Submitter rationale: The ATM c.931A>G (p.I311V) variant has been reported in heterozygosity in at least two individuals with breast cancer (PMID: 27913932, 33471991). It was observed in 2/34538 chromosomes of the Latino subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID 229990). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
