Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.3806G>C (p.Cys1269Ser), citing Ambry Variant Classification Scheme 2023: The p.C1269S variant (also known as c.3806G>C), located in coding exon 9 of the MSH6 gene, results from a G to C substitution at nucleotide position 3806. The cysteine at codon 1269 is replaced by serine, an amino acid with dissimilar properties. This variant was reported in 2/60,466 breast cancer cases and in 1/53,461 controls (Dorling et al. N Engl J Med. 2021 02;384:428-439). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
