Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000059.4(BRCA2):c.7606T>C (p.Ser2536Pro), citing ACMG Guidelines, 2015. This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 7606, where T is replaced by C; at the protein level this means replaces serine at residue 2536 with proline — a missense variant. Submitter rationale: This missense variant replaces serine with proline at codon 2536 of the BRCA2 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in a homozygous state in individuals with a personal or family history of breast and/or ovarian cancer (PMID: 23635950). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
