Uncertain significance — the classification assigned by Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine to NM_144991.3(TSPEAR):c.1079A>G (p.Lys360Arg), citing LMM Criteria: The p.Lys360Arg variant in TSPEAR has not been previously reported in individual s with hearing loss, but has been identified in 1/66726 of European chromosomes by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org; rs375 829414). The lysine (Lys) at position 360 is not conserved in mammals or evoluti onary distant species with rabbit and armadillo having an arginine (Arg), raisin g the possibility that a change at this position may be tolerated. Additional co mputational prediction tools do not provide strong support for or against an imp act to the protein. In summary, the clinical significance of the p.Lys360Arg var iant is uncertain.
