Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_015559.3(SETBP1):c.931C>A (p.Leu311Ile), citing Ambry General Variant Classification Scheme_2022: The c.931C>A (p.L311I) alteration is located in exon 4 (coding exon 3) of the SETBP1 gene. This alteration results from a C to A substitution at nucleotide position 931, causing the leucine (L) at amino acid position 311 to be replaced by an isoleucine (I). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is well conserved in available vertebrate species. This alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
