NM_004415.4(DSP):c.6062C>G (p.Ala2021Gly) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 6062, where C is replaced by G; at the protein level this means replaces alanine at residue 2021 with glycine — a missense variant. Submitter rationale: The p.A2021G variant (also known as c.6062C>G), located in coding exon 24 of the DSP gene, results from a C to G substitution at nucleotide position 6062. The alanine at codon 2021 is replaced by glycine, an amino acid with similar properties. This alteration has been reported in a control cohort (Mazzarotto F et al. Circulation, 2020 Feb;141:387-398). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
