NM_144508.5(KNL1):c.1813A>G (p.Ser605Gly) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1891A>G (p.S631G) alteration is located in coding exon 10 of the KNL1 gene. This alteration results from a A to G substitution at nucleotide position 1891, causing the serine (S) at amino acid position 631 to be replaced by a glycine (G). This allele was reported in one heterozygous individual in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is not well conserved in available vertebrate species. This alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
