Submitter rationale: The c.1271C>T (p.T424I) alteration is located in exon 8 (coding exon 8) of the COL5A1 gene. This alteration results from a C to T substitution at nucleotide position 1271, causing the threonine (T) at amino acid position 424 to be replaced by an isoleucine (I). Based on data from the Genome Aggregation Database (gnomAD) database, the COL5A1 c.1271C>T alteration was observed in 0.0008% (2/248,984) of total alleles studied, with a frequency of 0.002% (2/111,338) in the European (non-Finnish) subpopulation.
