Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003791.4(MBTPS1):c.106C>G (p.Pro36Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 36 of the MBTPS1 protein (p.Pro36Ala). This variant is present in population databases (rs138570962, gnomAD 0.06%). This variant has not been reported in the literature in individuals affected with MBTPS1-related conditions. ClinVar contains an entry for this variant (Variation ID: 2229015). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
