Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004415.4(DSP):c.3134G>A (p.Arg1045Gln), citing Ambry Variant Classification Scheme 2023. This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 3134, where G is replaced by A; at the protein level this means replaces arginine at residue 1045 with glutamine — a missense variant. Submitter rationale: The p.R1045Q variant (also known as c.3134G>A), located in coding exon 23 of the DSP gene, results from a G to A substitution at nucleotide position 3134. The arginine at codon 1045 is replaced by glutamine, an amino acid with highly similar properties. This variant has been reported in a dilated cardiomyopathy (DCM) cohort (Mazzarotto F et al. Circulation, 2020 Feb;141:387-398). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
