Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.533G>A (p.Gly178Glu), citing Ambry Variant Classification Scheme 2023: The p.G178E variant (also known as c.533G>A), located in coding exon 3 of the CHEK2 gene, results from a G to A substitution at nucleotide position 533. The glycine at codon 178 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
