This variant lies in the NF1 gene (transcript NM_001042492.3) at coding-DNA position 2447, where G is replaced by A; at the protein level this means replaces arginine at residue 816 with glutamine — a missense variant. Submitter rationale: The p.R816Q variant (also known as c.2447G>A), located in coding exon 21 of the NF1 gene, results from a G to A substitution at nucleotide position 2447. The arginine at codon 816 is replaced by glutamine, an amino acid with highly similar properties. This alteration was not observed in unselected male breast cancer patients and was observed with an allele frequency of 0.0001 in 12490 male controls of Japanese ancestry (Momozawa Y et al. Nat Commun, 2018 10;9:4083). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
