NM_007194.3(CHEK2):c.684-?_*154+?dup was classified as Uncertain significance for Familial cancer of breast by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross duplication of the genomic region encompassing exons 6-15 of the CHEK2 gene. The 5' boundary is likely confined to the intronic region between exons 5 and 6. The 3' end of this event is unknown as it extends through the termination codon beyond the assayed region for this gene and may encompass additional genes. Gross duplications of CHEK2 that include the termination codon have not been reported in the literature. This assay is unable to determine the genomic location of the duplicated region(s), and the functional significance of a copy number gain encompassing these exons has not been determined. In summary, this is a novel terminal duplication with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance.
