Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_000251.3(MSH2):c.2525A>T (p.Glu842Val), citing Sema4 Curation Guidelines: The MSH2 c.2525A>T (p.E842V) variant has not been reported in the literature to our knowledge. This variant was observed in 2/8712 chromosomes in the African/African American population, according to the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID: 220488). In silico tools suggest the impact of the variant on protein function is deleterious, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
