Submitter rationale: This sequence change replaces valine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 253 of the ATL1 protein (p.Val253Ile). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with autosomal dominant hereditary spastic paraplegia (PMID: 15596607, 16401858, 17285536, 20932283, 24604904, 26208798).
