Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006005.3(WFS1):c.1628T>G (p.Leu543Arg), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This missense change has been observed in individual(s) with Wolfram syndrome (PMID: 12754709, 16151413, 32179840). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is present in population databases (no rsID available, gnomAD 0.0009%). This sequence change replaces leucine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 543 of the WFS1 protein (p.Leu543Arg).
