This variant lies in the WFS1 gene (transcript NM_006005.3) at coding-DNA position 1456, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 486 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This premature translational stop signal has been observed in individual(s) with autosomal recessive optic atrophy and/or autosomal recessive Wolfram syndrome (PMID: 12754709, 33841295). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. For these reasons, this variant has been classified as Pathogenic. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Gln486*) in the WFS1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 405 amino acid(s) of the WFS1 protein.
