Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.7269A>T (p.Glu2423Asp), citing Ambry Variant Classification Scheme 2023: The p.E2423D variant (also known as c.7269A>T), located in coding exon 48 of the ATM gene, results from an A to T substitution at nucleotide position 7269. The glutamic acid at codon 2423 is replaced by aspartic acid, an amino acid with highly similar properties. This variant was detected once in a cohort of 1663 Brazilian breast cancer patients who underwent hereditary multigene panel testing (Guindalini RSC et al. Sci Rep, 2022 Mar;12:4190). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
