Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000260.4(MYO7A):c.3988G>A (p.Ala1330Thr), citing LabCorp Variant Classification Summary - May 2015: Variant summary: MYO7A c.3988G>A (p.Ala1330Thr) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 1.2e-05 in 248430 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3988G>A has been observed in at-least one heterozygous individual with congenital bilateral hearing loss, without strong evidence of causality (example: Florentine_2022). These report(s) do not provide unequivocal conclusions about association of the variant with Usher Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
