Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_017617.5(NOTCH1):c.721C>T (p.His241Tyr), citing Ambry Variant Classification Scheme 2023: The p.H241Y variant (also known as c.721C>T), located in coding exon 4 of the NOTCH1 gene, results from a C to T substitution at nucleotide position 721. The histidine at codon 241 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
