Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024120.5(NDUFAF5):c.33T>A (p.Cys11Ter), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with NDUFAF5-related conditions. This sequence change creates a premature translational stop signal (p.Cys11*) in the NDUFAF5 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in NDUFAF5 are known to be pathogenic (PMID: 26275793, 30473481, 32918965). This variant is not present in population databases (gnomAD no frequency). For these reasons, this variant has been classified as Pathogenic.
