NM_000393.5(COL5A2):c.3002G>A (p.Arg1001His) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1001H variant (also known as c.3002G>A), located in coding exon 43 of the COL5A2 gene, results from a G to A substitution at nucleotide position 3002. The arginine at codon 1001 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
