Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 0.00014 in 251288 control chromosomes. This frequency is not significantly higher than estimated for disease-causing variants in VPS13D, allowing no conclusion about variant significance. c.3161A>C has been observed in a homozygous individual affected with Spinocerebellar Ataxia, Autosomal Recessive 4 (Bildirici_2023) as well as in an interal patient who was presumed compound heterozygous with clinical features of Spinocerebellar Ataxia, Autosomal Recessive 4. These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
