Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.9907A>G (p.Ile3303Val), citing Ambry Variant Classification Scheme 2023: The p.I3303V variant (also known as c.9907A>G), located in coding exon 60 of the DNAH11 gene, results from an A to G substitution at nucleotide position 9907. The isoleucine at codon 3303 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
