Uncertain significance for Charcot-Marie-Tooth disease type 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_030962.4(SBF2):c.402+11_402+21del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SBF2 gene (transcript NM_030962.4) at 11 bases into the intron immediately after coding-DNA position 402 through 21 bases into the intron immediately after coding-DNA position 402, deleting this region. Submitter rationale: This sequence change falls in intron 4 of the SBF2 gene. It does not directly change the encoded amino acid sequence of the SBF2 protein. This variant is present in population databases (no rsID available, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with SBF2-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
