Uncertain Significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by All of Us Research Program, National Institutes of Health to NM_001613.4(ACTA2):c.132G>T (p.Gly44=), citing ACMG Guidelines, 2015. This variant lies in the ACTA2 gene (transcript NM_001613.4) at coding-DNA position 132, where G is replaced by T; at the protein level this means the protein sequence is unchanged (glycine at residue 44 retained) — a synonymous variant. Submitter rationale: This variant is located in the ACTA2 protein. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with ACTA2-related disorders in the literature. This variant has been identified in 1/251062 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
