NM_001370259.2(MEN1):c.640G>A (p.Gly214Ser) was classified as Uncertain Significance for Multiple endocrine neoplasia, type 1 by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the MEN1 gene (transcript NM_001370259.2) at coding-DNA position 640, where G is replaced by A; at the protein level this means replaces glycine at residue 214 with serine — a missense variant. Submitter rationale: This missense variant replaces glycine with serine at codon 214 of the MEN1 protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in two individuals in the MEN1 Universal Mutation Database (PMID: 12112656; http://www.umd.be/MEN1/). This variant has been identified in 6/251016 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
