Uncertain significance for Mucopolysaccharidosis type 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000046.5(ARSB):c.917T>C (p.Leu306Ser), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ARSB gene (transcript NM_000046.5) at coding-DNA position 917, where T is replaced by C; at the protein level this means replaces leucine at residue 306 with serine — a missense variant. Submitter rationale: This sequence change replaces leucine with serine at codon 306 of the ARSB protein (p.Leu306Ser). The leucine residue is moderately conserved and there is a large physicochemical difference between leucine and serine. This variant is present in population databases (rs143111531, ExAC 0.006%). This variant has not been reported in the literature in individuals affected with ARSB-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
