Uncertain significance for Charcot-Marie-Tooth disease axonal type 2P — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001005373.4(LRSAM1):c.929T>G (p.Leu310Arg), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with LRSAM1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant is present in population databases (no rsID available, gnomAD 0.009%). This sequence change replaces leucine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 310 of the LRSAM1 protein (p.Leu310Arg).
