Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.6293A>G (p.Asn2098Ser), citing Ambry Variant Classification Scheme 2023: The p.N2098S variant (also known as c.6293A>G), located in coding exon 45 of the POLE gene, results from an A to G substitution at nucleotide position 6293. The asparagine at codon 2098 is replaced by serine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
