Uncertain significance for Immunodeficiency 25 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_198053.3(CD247):c.488C>G (p.Pro163Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with arginine, which is basic and polar, at codon 163 of the CD247 protein (p.Pro163Arg). This variant is present in population databases (rs376046446, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with CD247-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
