Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_017654.4(SAMD9):c.3361G>T (p.Asp1121Tyr), citing Ambry Variant Classification Scheme 2023: The p.D1121Y variant (also known as c.3361G>T), located in coding exon 1 of the SAMD9 gene, results from a G to T substitution at nucleotide position 3361. The aspartic acid at codon 1121 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
