This variant lies in the ADAMTS2 gene (transcript NM_014244.5) at coding-DNA position 2569, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 857 with lysine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid with lysine at codon 857 of the ADAMTS2 protein (p.Glu857Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine. This variant is present in population databases (rs747653130, ExAC 0.006%). This variant has not been reported in the literature in individuals with ADAMTS2-related conditions.
