This variant lies in the MSH6 gene (transcript NM_000179.3) at coding-DNA position 2018, where C is replaced by T; at the protein level this means replaces proline at residue 673 with leucine — a missense variant. Submitter rationale: Variant summary: MSH6 c.2018C>T (p.Pro673Leu) results in a non-conservative amino acid change located in the DNA mismatch repair protein MutS, connector domain (IPR007860) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 250906 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2018C>T in individuals affected with Hereditary Non-Polyposis Colon Cancer and no experimental evidence demonstrating its impact on protein function have been reported.
