Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001283009.2(RTEL1):c.3014A>G (p.Lys1005Arg), citing Ambry Variant Classification Scheme 2023: The p.K1005R variant (also known as c.3014A>G), located in coding exon 30 of the RTEL1 gene, results from an A to G substitution at nucleotide position 3014. The lysine at codon 1005 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
