NM_000093.5(COL5A1):c.431_432delinsAT (p.Thr144Asn) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.431_432delCGinsAT variant (also known as p.T144N), located in coding exon 3 of the COL5A1 gene, results from an in-frame deletion of CG and insertion of AT at nucleotide positions 431 to 432. This results in the substitution of the threonine residue for an asparagine residue at codon 144, an amino acid with similar properties. Based on data from gnomAD, this allele has an overall frequency of 0.0056% (14/250192) total alleles studied. The highest observed frequency was 0.0145% (5/34580) of Latino alleles. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
