Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_174934.4(SCN4B):c.133A>C (p.Asn45His), citing Ambry Variant Classification Scheme 2023: The p.N45H variant (also known as c.133A>C), located in coding exon 2 of the SCN4B gene, results from an A to C substitution at nucleotide position 133. The asparagine at codon 45 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
